The university's first classes in 1935 were attended by 85 students. Many of these students viewed education as a means to enhance their employment opportunities during the Great Depression.
The first director of the university was Mr. Samuel Fischer Scott, an alumnus of Virginia Union University. His primary focus was on maintaining the solvency of the school. Dr. Lyman Beecher Brooks, also a Virginia Union alumnus, succeeded Mr. Scott as director in 1938. Dr. Brooks went on to serve as provost from 1963 to 1969 and then as the university's first president from 1969 to 1975.
In 1942, the school became independent of Virginia Union University and was renamed Norfolk Polytechnic College. Within two years, the Virginia Legislature made it a part of Virginia State College (now Virginia State University). By 1950, the college had nearly 1,000 students and 50 faculty members. In 1952, the college's athletic teams adopted the name and identity of the "Spartans."
The university has continued to grow and expand over the years. By 1969, it had separated from Virginia State College again and was renamed Norfolk State College. That year, the college was accredited by the Southern Association of Colleges and Schools and had an enrollment of 5,400 students, making it one of the larger historically black colleges and universities (HBCUs) in the nation. In 1979, the college gained university status and became known as Norfolk State University.
